Clostridium difficile Cytotoxicity

Clostridium difficile toxin is a common cause of antibiotic associated diarrhea (AAD) and pseudomembranous colitis (PMC). The toxigenic culture method in which organisms are cultured and the stool is tested by the Cell Culture Cytotoxicity Neutralization assay (CCNA) remains the diagnostic “gold standard” for detection of toxigenic C. difficile. The CCNA consists of two reactions—detection of toxin and toxin neutralization by the C. difficile antitoxin. The method is extremely sensitive and can detect toxin levels as low as 1 picogram. This testing should be performed using stool specimen filtrate, however our evaluation shows that when a pure isolate of C. difficile is used as an alternative specimen, the sensitivity of the test is higher than the raw stool specimen. A delay in transport of unpreserved stool samples can lead to degradation of toxin in samples leading to a false negative result. CCNA testing of C. difficile isolates from preserved stool samples (enriched in cooked meat broth or thioglycolate broth) provides a higher level of sensitivity than that obtained with direct testing of unpreserved stool samples.Clostridium difficile Cytotoxicity Assay - Clostridium difficile is a spore-forming bacteria that is part of the normal intestinal flora. With intestinal overgrowth, c. Difficile is the major cause of antibiotic-associated diarrhea and colitis. C. Difficile is the primary cause of pseudomembranous colitis. The organism produces cytotoxins that lead to frequent, foul-smelling, watery stools.

What is Clostridium

Clostridium, genus of rod-shaped, usually gram-positive bacteria, members of which are found in soil, water, and the intestinal tracts of humans and other animals. Most species grow only in the complete absence of oxygen. Dormant cells are highly resistant to heat, desiccation, and toxic chemicals and detergents. The species are variable in size. A typical species, C. butyricum, ranges from 0.6 micrometre across by 3 to 7 micrometres long. The toxins produced by C. botulinum, the causative agent of botulism, are the most potent poisons known. The toxin of C. tetani causes tetanus when introduced into damaged or dead tissue. C. perfringens, C. novyi, and C. septicum can cause gangrene in humans. Other forms of acute clostridial infection commonly occur in livestock and waterfowl.Clostridium perfringens is one of the most common types of foodborne illness in the United States. CDC estimates it causes nearly 1 million cases of foodborne illness each year.C. perfringens is commonly found on raw meat and poultry. It prefers to grow in conditions with very little or no oxygen, and under ideal conditions can multiply very rapidly. Some strains of C. perfringens produce a toxin in the intestine that causes illness.

What is Cytotoxicity?

In a nutshell, cell cytotoxicity refers to the ability of certain chemicals or mediator cells to destroy living cells. By using a cytotoxic compound, healthy living cells can either be induced to undergo necrosis (accidental cell death) or apoptosis (programmed cell death).Given this information, the ability to accurately measure cytotoxicity can prove to be a very valuable tool in identifying compounds that might pose certain health risks in humans. This can be of vital importance during the research phase of developing new pharmaceutical products to ensure the safety of the end-users.Cell cytotoxicity, measure cell cytotoxicityMeasuring cell cytotoxicity also proves to be quite indispensable in the process of developing therapeutic anti-cancer drugs. By determining the cytotoxicity levels of cancer cells, anti-cancer medications can hinder the proliferation of target cells either by messing with their genetic material or by blocking the nutrients that the cells needs to survive.Additionally, understanding the mechanisms involved in cytotoxicity can likewise give researchers a more in-depth knowledge on the biological processes (both normal and abnormal) governing cell growth, cell proliferation and death.Looking for "how to lower hematocrit on trt?".
